If your employer makes you sign a handbook saying you won't possess a firearm while under employment, does signing mean that you voluntarily forfit your 2nd amendment rights?

Legally they can't dictate whether an employee owns firearms. They can say that you can't have them while working or on company property but no company on American soil can strip any American citizen of their rights guaranteed by the Constitution.

So if the contract is signed and then you are fired because of owning and operating firearms on free time off company property, then you have legal grounds to sue for violation of constitutional rights? GL4x4 legal advice

Don't assume their intentions. If it's that vague, that means they can interpret it in any way they choose if the need (in their eyes) calls for it.
