Lock them when you plan on using 4wd, and unlock them during normal pavement driving. All it does is disengages the axle shaft to the hub.

If you have them unlocked, and you put it in 4wd, the front axle shafts will spin, but they will not provide power to the tires.

Depending on your transfer case, (if you can put the front in neutral) you can save a lot of wear/tear on your drive train by leaving the t-case in front neutral, and having the hubs unlocked. This means the front drives haft won't be spinning while you are driving.

Also, carry a spare, and know how to change them. The cheapo milemarkers and entry level warn lockouts are basically the fuse up front when you're beating up on the front axle. They break before anything else.
